The dynamic response of an oscillating microcantilever with a gold-coated tip interacting with dissimilar functionalized silica surfaces was studied in electrolyte solutions with pH ranging from 4 to 9. Silica surfaces were chemically modified, yielding dissimilar surfaces with -Br, -NH(2), and -CH(3) functional group terminations. The relative hydrophobicity of the surfaces was characterized by contact angle measurements. The surface charge of the functionalized surfaces was first probed with commonly used static AFM measurements and serves as a reference to the dynamic response data. The amplitude and phase of the cantilever oscillation were monitored and used to calculate the effective interaction stiffness and damping coefficient, which relate to the electrical double layer interactions and also to distance-dependent hydrodynamic damping at the solid/water interface. The data for the dynamic response of the AFM over silica surfaces as a function of chemical functionalization and electrolyte pH show that the effective stiffness has a distinctive dependence on the surface charge of functionalized silica surfaces. The hydrodynamic damping also correlates strongly with the relative hydrophobicity of the surface. The data reported here indicate that interfacial properties can be strongly affected by changing the chemical composition of surfaces.  相似文献

Photonic crystals fabricated from the colloidal spheres of polymethyl methacrylate (PMMA) using the inward-growing self-assembly technique were subsequently in-filled with zinc oxide (ZnO) prepared by the sol-gel process. The polymer template was removed by heat treatment and chemical method to get ZnO-inverse photonic crystal. The structural quality of the inverse photonic crystal obtained by the chemical method was found to be superior to that obtained by heat treatment. The ZnO-inverse photonic crystal obtained by the chemical method was further treated at an elevated temperature to ensure the crystalline nature of ZnO. Laser-induced emission studies on ZnO-inverse photonic crystals were carried out at an excitation wavelength of 325 nm. The emission spectra showing UV and visible bands at collection angle of 45° from the direction of excitation helped to establish the role of crystalline ZnO.  相似文献

Many aspects of photophysical, photostability and laser properties of kiton red S dye remain unresolved, particularly for pumping with 578 nm radiation of CVL and 532 nm output of Nd:YAG lasers, and these are studied using different alcohol- and water-based binary solvents. Our results show that methanol is better suited for low and high-repetition-rate KRS dye lasers because of its superior laser efficiency, photostability as well as photothermal properties.  相似文献

Steady-state fluorescence measurements and isothermal titration calorimetric experiments have been performed to study the interaction between a telechelic polymer, pyrene-end-capped poly(ethylene oxide) (PYPY), and sodium alkyl sulfate surfactants having decyl, dodecyl, and tetradecyl hydrocarbon tails. Fluorometric results suggest polymer-surfactant interaction in the very low range of polymer concentrations. The relative variation in the excimer to monomer pyrene emission intensities with varying surfactant concentration reveals that initial addition of surfactant favors intramolecular preassociation until the surfactant molecules start binding with the ethylene oxide (EO) chain. With the growing number of surfactant aggregates along the EO chain, the association becomes hindered due to the polyelectrolyte effect. The results from microcalorimetric titrations in the low concentration range of PYPY solution (approximately 10(-6) M) with alkyl sulfates suggest two kinds of surfactant-polymer interactions, one with the polymer hydrophobic end groups and the other with the ethylene oxide backbone. The overall polymer-surfactant interaction starts at a much lower surfactant concentration for the hydrophobically modified polymers compared to that in the case of unsubstituted poly(ethylene oxide) homopolymer. From the experiments critical aggregation concentration values and the second critical concentration where free micelles start forming have been determined. An endeavor has been made to unveil the mechanism underlying the corresponding associations of the surfactants with the polymer.  相似文献

The glycoprotein BclA is an important constituent of the exosporium of Bacillus anthracis spores. This glycoprotein is substituted with an oligosaccharide composed of a beta-L-rhamnoside substituted with the previously unknown terminal saccharide, 2-O-methyl-4-(3-hydroxy-3-methylbutanamido)-4,6-dideoxy-D-glucopyranose, also referred to as anthrose. Anthrose has not been found in spores of B. cereus and B. thuringiensis, making it a potential species-specific marker for B. anthracis. In order to study the antigenicity of anthrose, efficient syntheses of an anthrose-containing trisaccharide and a series of structurally related analogues were developed. The analogues lacked either the methyl ether at C-2 or contained modified C-4 amino functionalities of anthrose. The synthetic compounds were equipped with an aminopropyl spacer to facilitate conjugation to the carrier proteins mariculture Keyhole Limpet Hemocyanin (mcKLH) and bovine serum albumin (BSA). Serum antibodies of rabbits immunized with live or irradiated spores of B. anthracis Sterne 34F(2) were able to recognize the synthetic trisaccharide-mcKLH conjugate. The specificity of the interaction was confirmed by competitive inhibition with the free- and BSA-conjugated trisaccharides. Inhibition using the trisaccharide analogues demonstrated that the isovaleric acid moiety of anthrose is an important structural motif for antibody recognition. These data demonstrate that 1) anthrose is a specific antigenic determinant of the B. anthracis Sterne spore; 2) this antigen is presented to the immune system of rabbits receiving the anthrax live-spore vaccine; 3) synthetic analogues of the oligosaccharide retain the antigenic structure; and 4) the antigenic region is localized to specific terminal groups of the oligosaccharide. Collectively these data provide an important proof-of-concept step in the synthesis and development of spore-specific reagents for detection and targeting of non-protein structures in B. anthracis.  相似文献

The binding of chlorin p6, a photosensitizer having basic tetrapyrrole structure, to bovine serum albumin (BSA) and oxidation of the protein following photodynamic treatment is studied. The Stern-Volmer plot indicates that binding of chlorin p6 to BSA was of single class. Binding parameters, binding association constant and number of binding sites, were found to be 1.62+/-0.27 x 10(5)M(-1) and 1.086+/-.019, respectively. Photodynamic oxidation of protein was studied by (i) loss of intrinsic fluorescence of protein, (ii) protein carbonyl formation, (iii) protein hydroperoxide (iv) formation of TCA soluble amino groups and (v) SDS-polyacrylamide gel electrophoresis (SDS-PAGE). Intrinsic protein fluorescence was observed to decrease almost linearly as a function of irradiation time at a fixed concentration of chlorin p6 and with increasing concentration of chlorin p6 at fixed time of irradiation. Protein carbonyl and hydroperoxide formation was found to increase with increasing photodynamic treatment. No significant increase in 5% TCA soluble amino groups was observed. SDS-polyacrylamide gel electrophoresis (SDS-PAGE) reveals that photodynamic treatment of BSA in presence of chlorin p6, rose bengal and riboflavin causes non-specific fragmentation of protein. Photodynamic carbonyl formation by chlorin p6 was not inhibited by sodium formate (100 mM) or mannitol (25 mM) but was significantly inhibited by sodium azide (2 mM). Protein carbonyl formation increased almost 90% when H2O was replaced by D2O. The results show that chlorin p6 induced photodynamic oxidation of BSA was mainly mediated by singlet oxygen.  相似文献

In continuation of our recent study on the steady state photophysics of a biologically active beta-carboline derivative, 3-acetyl-4-oxo-6,7-dihydro-12H indolo-[2,3-a] quinolizine (AODIQ), in the present article we have investigated the effect of nanocavity confinement on the excited state dynamics and rotational relaxation of the probe using picosecond time resolved fluorescence and fluorescence anisotropy techniques. The polarity dependent intramolecular charge transfer process is responsible for the remarkable sensitivity of this biological fluorophore in micellar environments. The fluorescence anisotropy decay of AODIQ incorporated inside the micelle is biexponential. The rotational motion of the probe was interpreted on the basis of a two step model consisting of a fast restricted rotation of the probe and a slow lateral diffusion of the probe in the micelle; both coupled to the overall rotation of the micelle. Experimental results reveal that micellar environment causes significant retardation of both the wobbling as well as the translational motion of the probe.  相似文献

In methanol, the reaction of Cu(ClO(4))(2).6H(2)O and a sterically constrained piperazine imine phenol ligand (H(2)L), in the presence of NEt(3), affords a novel tetranuclear copper(II) complex of formula [Cu(II)(4)(mu(3)-L)(2)(mu-OH)(2)(H(2)O)(2)](ClO(4))(2).H(2)O (1). The X-ray structure of this complex shows an elongated Cu(4) quasi-tetrahedron coordinated to two hexadentate chair-(e,a)-mu(3)-piperazine bridging ligands. Variable-temperature magnetic studies show an S(t) = 0 spin ground state resulting from antiferromagnetic interactions between Cu(II) ions within the complex.  相似文献

Some mixed-ligand thorium(IV) complexes with the general formula [Th(OOCCH3)4?nLn] (L = anions of myristic, palmitic or stearic acid and n = 1–4) have been synthesized by the stepwise substitution of acetate ions of thorium(IV) acetate with straight chain carboxylic acids in toluene under reflux. The complexes were characterized by elemental analyses, spectral (electronic, infrared, NMR and powder XRD) studies, electrical conductance and magnetic susceptibility measurements. Doubly and triply bridged coordination modes of the ligands were established by their infrared spectra and nano-size of the complexes by powder XRD. Room temperature magnetic susceptibility measurements revealed diamagnetic nature of the complexes. Electronic absorption spectra of the complexes showed π  π*, n  π* and charge transfer transitions. Molar conductance values indicated the complex to be non-electrolytes. These are a new type of mixed-ligand thorium(IV) complexes for which a nano-sized, oxygen bridged polymeric structure has been established on the basis of physico-chemical studies.  相似文献

Polymerizable phospholipids are organic monomers that are able to self-assemble into membranes. This is quite similar to the self assembly of naturally occurring lipid membranes found in biological systems. The polymerizable lipids can then be polymerized to stabilize these membranes. These stabilized structures have a number of different morphologies which offer a variety of technically attractive features. This article reviews the chemical and physical properties of these materials in terms of their technological applications.  相似文献
